Anticancer activity of oleanolic acid and its derivatives: Recent advances in evidence, target profiling and mechanisms of action.

Systematic review

This paper reviews the preclinical studies, literature evidence, target analysis, and anticancer mechanisms of oleanolic acid (OA) and its derivatives. It discusses the potential of OA and its derivatives as anticancer drugs, highlighting their effects on cancer cell proliferation, apoptosis, autophagy, and other mechanisms.

The effect of a toothpaste containing oleanolic acid in reducing plaque and gingivitis: a proof-of-concept randomized trial.

RCTn = 99

This proof-of-concept randomized controlled trial assessed the safety and efficacy of an oleanolic acid-containing toothpaste in reducing gingival inflammation and plaque in 99 patients with gingivitis. After two weeks, all groups showed significant reductions in clinical parameters, but the oleanolic acid toothpaste did not provide superior antiplaque and antigingivitis effects compared to fluoride toothpaste.

Pharmacokinetics of maslinic and oleanolic acids from olive oil - Effects on endothelial function in healthy adults. A randomized, controlled, dose-response study.

RCTHeart Health

Randomized, controlled, dose-response study assessing pharmacokinetics of maslinic and oleanolic acids from olive oil in healthy adults. Maslinic acid showed greater bioavailability than oleanolic acid, and consumption of these triterpenes was associated with improved endothelial function.

Bioavailability and systemic transport of oleanolic acid in humans, formulated as a functional olive oil.

RCTn = 22

The BIO-OLTRAD trial is a double-blind, randomized controlled study with 22 participants receiving a single dose of 30 mg oleanolic acid formulated as a functional olive oil. The study measured the bioavailability of oleanolic acid, revealing maximum serum concentrations and associations with serum albumin and triglyceride-rich lipoproteins.

Prevention of type 2 diabetes in prediabetic patients by using functional olive oil enriched in oleanolic acid: The PREDIABOLE study, a randomized controlled trial.

RCTn = 176Nutrition

The PREDIABOLE study is an RCT assessing the effect of oleanolic acid-enriched olive oil on diabetes prevention in prediabetic individuals. The intervention group showed a reduced risk of developing type 2 diabetes compared to the control group, with a hazard ratio of 0.45.

Lipid-lowering effects of oleanolic acid in hyperlipidemic patients.

RCTHeart Health

Small-scale clinical trial evaluating the hypolipidemic effect of oleanolic acid (OA) in hyperlipidemic patients. OA administration for four weeks significantly decreased TC, TG, and HDLC levels in serum, and differentially regulated gene expression involved in lipid metabolism.

Therapeutic efficacy of L-ornithine-L-aspartate infusions in patients with cirrhosis and hepatic encephalopathy: results of a placebo-controlled, double-blind study.

RCTn = 126Brain Health

RCT of 126 patients with cirrhosis and hepatic encephalopathy comparing L-ornithine-L-aspartate (OA) to placebo. OA treatment improved NCT-A performance times, postprandial venous ammonia concentrations, mental state gradation, and PSEI compared to placebo. Mild gastrointestinal disturbances were observed in 5% of OA-treated patients.

Randomized, double-blind, placebo-controlled, consumer rechallenge test of Olean salted snacks.

RCTn = 57

A randomized, double-blind, placebo-controlled, crossover rechallenge study compared gastrointestinal symptoms after ingestion of Olean (olestra) chips versus triglyceride chips in 57 subjects. No significant difference in gastrointestinal symptoms was found between the two groups, suggesting previous symptoms attributed to Olean may have been misattributed.

Integrating network pharmacology and animal experimental validation to investigate the action mechanism of oleanolic acid in obesity

Animal studyPhysique

The study investigates the mechanism of oleanolic acid (OA) in treating obesity using network pharmacology and animal models. It identifies key targets and pathways, including PPARG and PPAR signaling, and validates OA's binding potential and stability with these targets, suggesting its potential as an anti-obesity treatment.

Effect of the moderate consumption of olestra in patients receiving long-term warfarin therapy.

RCTn = 40

RCT evaluating the effect of moderate consumption of snacks containing olestra on INR in 40 patients on long-term warfarin therapy. No significant effect on INR was observed after 1 or 2 weeks. Gastrointestinal side effects were reported in both groups.

Oleanolic acid improves pulmonary morphofunctional parameters in experimental sepsis by modulating oxidative and apoptotic processes.

Animal studyn = 78Inflammation

Animal study comparing oleanolic acid (OA) and dexamethasone on lung mechanics, histology, inflammation, and apoptosis in experimental sepsis. Both OA and DEXA improved lung mechanics and histology, with OA showing distinct anti-inflammatory mechanisms such as lower inducible nitric oxide synthase expression and higher superoxide dismutase.

[Treatment of actinic keratoses with birch bark extract: a pilot study].

Studyn = 28Skin Hair Health

Pilot study of birch bark ointment in 28 patients with actinic keratoses. Monotherapy cleared more than 75% of lesions in 79% of patients, while combination with cryotherapy had a 93% response rate. The treatment was well tolerated and effective.
